脱盐乳清粉代谢产物的抗氧化

摘要:比较研究以脱盐乳清粉为主要原料,采用乳杆菌A-2进行发酵制备的代谢产物(LF),并初步探
讨了工抗氧化和抗肿活性的功能。通过对发酵液的pH、总糖和Ca2含量的测定,筛选出最佳菌悬液浓度
为1oml-用ABTS法和ORAC法来測定和评价乳杆菌A-2发酵产物的抗氧化作用。在反应体系中,脱盐
乳清粉和IF的ABTS清除率的IC50分别为34.29mgmL和19.74mgmL,LF的ABTS清除率的能力优于脱盐
乳清粉;其相对ORAC值分别为75.461 umol Trolox/g和114.78 umol Trolox/g,LF清除AAPH的能力为脱盐乳
清粉的1.5倍,结果显示LF具有一定抗氧化活性。采用MTT法检测LF对人舌癌细胞系Ca-27细胞增殖的
彩响,LF的ICS0为19.1mgmL,且浓度在4-32mg/mL的范围内,对人舌癌细胞系Cal-27细胞的抑制率
呈剂量依赖性,与脱盐乳清粉组比较,差异具有显著性。

Abstract: In this study, the desalted whey powder was used as the principal raw material, using Lactobacillus A
for fermentation, to produce antioxidant activity and antitumor activity metabolite(LF). By measuring PH valu
of fermentation bacteria, total sugar and Ca"concentration, the best concentration of the bacterial suspension is
determined as 1OD/m the antioxidant activity of Lactobacilus -2 whey fermentation product was evaluated b
ABTS and ORAC. In the reaction s stem, the ABTS free radical scavenging actiity C of desalted whey powder and
LF were 34. smg/ML and 1974m/ML respectively, the ABTS free radical scavenging activity of LE is better than
that of desalted whey powder; the ORAC assay of desalted whey powder and LF were 75.4 and 114. mol Trolox/
g rcspcctivcly. The results showed that LF had prominent antioxidant activity. Using MTT method to measure the
