    摘要:研究胁迫培养条件下,在对数生长期的雨生红球藻 Haematococcus pluvialis LUGU藻液中添加不同质量浓度
    叔丁基对羟基茴香醚( butyl hydroxyanisole,BHA)对其影响,并分别对细胞生物量、虾青素积累量以及八番茄红
    素合成酶psy基因表达量进行了测定。结果表明,BHA质量浓度为2mgL时,虾青素积累量显著提高,最大含量可
    达29.03mg/g,是对照组(14.30mgg)的2.03倍。而在相同的BHA诱导条件下,初始藻细胞的不同生长状态对细
    胞生长及虾青素合成也有很大影响:处于对数后期(14d)的细胞其最大虾青素含量可达293mg/g,分别是对数前
    期、中期和稳定期的2.93、1.01倍和1.73倍。实时荧光定量聚合酶链式反应分析显示,虾青素合成关键基因Dsy的表
    达受BHA的影响,当对数后期(14d)添加2mg/LBHA时,py基因相对表达水平为对照9.2倍。结果表明,适当质
    量浓度BHA不仅能够显著提高虾青素合成关键酶基因psy的表达水平,并且明显促进了微藻细胞内虾青素积累量
    为虾青素产业化提供了一种有效的策略。

    Effects of Butyl Hydroxyanisole on Astaxanthin Accumulation and the Transcriptional Expression Levels of
    hytoene Synthase(psy) Gene of Haematococcus pluvialis LUGU

    Abstract: Owing to its role as an important source for astaxanthin accumulation, Haematococcus pluvialis has gain
    more and more attention in recent years. In this research, we examined the effects of butyl hydroxyanisole(BHA)at
    different concentrations on the astaxanthin production under unfavorable conditions during the logarithmic growth phase of
    H. pluvialis UGU. The results demonstrated that 2 mg/L BHA was optimal for maximum astaxanthin production, which
    reached 29. 03 mg/, 2.03 times higher han that of the control(14.30 m /g). Moreover, under the same condition of BHA
    induction, initial algal cells with different ow h status also had a great influence on as axanthin synthesis. The accumulation
    of astaxanthin in the algal cells in the late exponential growth phase(14 d)reached the highest level of 29.3 mg/g, which
    was 2.93, 1. 01 and 1.73 times higher those in the early and middle exponential, and stationary growth phases, respectively
    IRT-PCR analysis shows that the transcriptional levels of phytoene synthase(psy)gene induced by 2 mg/L BHA was 9.2
    times higher that of control. These findings showed that appropriate concentration of BHA could not only increase the
    transcription of pygene, but also could obviously promote intracellular astaxanthin accumulation in microalgae, providing
    an effective strategy for the industrialization of astaxanthin
