Abstract Objective: To study the effect of enhanced 5-aminolevulinic acid-based photodynamic therapy
(ALA-PDT) by DMSO on A431 cytotoxicity in vitro and inhibition of mice skin squamous cell carcinoma( MSCC)in
vivo. Methods: A431 cells were co-cul ured with DMSO Apoptosis after ALA-PDT and protoporphyrin X(Pp)
concentration of A431 cells co-cul ue wh DMSO were detected by flow cytometry. The fluorescence intensity in
MSCC frozen sections was detected by laser scanning confocal microscopy. After ALA-PDT the MSCC suppress
rates were compared. Results: The Pp IX fluorescence intensity of 431 cells in the experimenta therapeutic group
howed DMSO dose dependant manner, was higher than that in the ALA alone or control group(P<0.01 ) After
ALA-PDT, the A431 cell apoptotic rates in the experimental roup were higher than that in the ALA alone or eontrol
group(P <0.01). The Ppx fluorescence intensity in the experimental group showed DMSO dose dependant manner
was higher than that in the AL A alone or control group (P <0. 01). After ALA-PDT the tumor suppressing rates
showed DMSO dose dependant manner in therapeutic group(P<0.05) Conclusion DMSO can promote ALA-PDI
effects on A431 cytotoxicity and MSCC inhibition

摘要】目的:研究DMSO参与5-氨基酮戊酸介导的光动力疗法(ALA- based photodynamic therapy,ALA
PDT)对A431细胞杀伤和小鼠皮肤鳞癌的抑制效应,探讨治疗皮肤肿瘤的优化作用。方法:DMS0千预后,采
用流式细胞仪检测A431细胞内PpX水平和PDT后细胞凋亡率。激光共聚焦显微镜检测小鼠鳞寤瘤体组织
的荧光强度,比较PDT后各组鳞癌小鼠的抑瘤率。结果:实验组A431细胞PpⅨ荧光强度率高于ALA组和空
白对照组(P<0.01),与DMSO含量呈浓度依赖性。PDT后实验组细胞凋亡率高于ALA组和空白对照组(P
<0.01)。实验组瘤体组织PpⅨ荧光强度值高于10%ALA对照组和10%DMSO对照组(P<0.01),与DMS0
含量呈浓度依赖性。实验组鱗癌小鼠PDT治疗后,抑瘤率高于对照组(P<0.05),并有量效依从关系。结
论:DMSO可促进ALA-PDT对A4311细胞杀伤和小鼠皮肤鳞癌的抑制效应。
