嗨比色法测定禽蛋中的胆固醇含量

摘要:目的建立晦比色法快速測定禽蛋中胆固醇含量的分析方法。方法以嗨比色法与分光光度计联用.
在波长505nm处直接测定经皂化、提取、浓缩和酶工作液催化后的禽蛋样品中胆固醇含量。结果本试验考
察了酶比色反应的最优显色温度(37℃)和显色时间(15min),并在最优试验条件下测得线性范固为0~2.0
mg/ml,曲线回归方程Y=0.3207X+0.0891,相关系数R为0.9990,检出限为1.0mg/100g,回收率达到
90.43%~98.12%,相对标准偏差小于2.45%。结论本方法操作简单、具有很好的精密度与准确度,适合鸡蛋
甲鸟貨、鹅蛋、鸽蛋等全蛋样品或蛋黄样品中胆固醇含量的快速检测。

ABSTRACT: Objective To establish a method for determination of cholesterol in eggs by enzyme colorimetry
Methods Determination of cholesterol in eggs by enzyme colorimetry coupled with spectrophotometer at 505 nm
aller samples wcrc prcparcd by saponification, cxtraction, conccntration and cnzymc calalyzed rcaction. Results
The optimal coloration Lcmpcraturc (37 C)and coloration timc(15 min )had bccn invcstigatcd. The mcthod had a
linear range of calibration curve of 0-2.0 mg/ml for determination of cholesterol in eggs, and the linear correlation
coefficients were more than 0.999. The detection limit were 1.0 mg/100 g and the relative standard deviations(RSD
were not more than 2. 45%. The recoveries for the spiked samples ranged from 90.43% to 98.12%. Conclusion This
method is simple and accurate, which is suitable for the determination of cholesterol in egg, duck egg, goose eg
pigeon cgg and other wholc cgg samples or cgg yolk samples
